Sensitivity of Escherichia coli to viral nucleic acid. XVI. Temperature conditions for Ca2+-dependent dna uptake in Escherichia coli.

چکیده
Properties of Ca2+- or Ba2+-dependent transfection and transformation in Escherichia coli were examined, using phi X174 replicative-form (RF) DNA and plasmid DNA. For the transfection and transformation, a heat pulse step was dispensable and the yield of transfectants was, in most E. coli strains, rather reduced by the heat treatment. The heat pulse step was also detrimental for the transformation of certain strains such as lipopolysaccharide mutants, The first stage of the DNA uptake process (formation of DNA . recipient cell complex) was dependent on low temperature and Ca2+ ion. A substantial amount of the complexed RF-DNA was released from the bacteria, by washing with a chilled Tris buffer. Although a RF-DNA . cell complex was formed even at 37 degree C or in chilled 0.05 m MgC12, the complex did not yield transfectants.

Cellular Morphology and Immunologic Properties of Escherichia coli Treated With Antimicrobial Antisense Peptide Nucleic Acid
Background & Objectives: Antisense peptide nucleic acids (PNA) that target growth essential genes show potent bactericidal properties without cell lysis. We considered the possibility that whether PNA treatment influence the bacteria total nucleic acids content and apply approach to develop a new delivery system to Dendritic cells (DCs). DCs are the most potent antigen presenting cells in th...

Background: The most important researches in molecular and genetic engineering fields are the finding of optimal methods for extraction of the genomic content of microorganisms and cells using them, can achieve the most amount and purity with the least time and cost. There are several methods for the extraction of bacterial DNA and the use of magnetic nanoparticles is one of the novel methods o...
متن کاملSensitivity of Escherichia coli to viral nucleic acid, XII. Ca2+- or Ba2+-facilitated transfection of cell envelope mutants.
Using various envelope mutants of Escherichia coli, the relationship between cell surface struc ture and the Ca2*or Ba2+-dependent competence for transfection was investigated. In contrast with rough strains, smooth bacteria treated with Ca2+ or Ba2+ were incompetent for the trans fection by 0 A RF. In E. coli K12 D21 derivatives, Ca2+-dependent competence remarkably in creased by IpsA l mut...
متن کاملEvaluation of the antibiotic resistance and prevalence of uropathogenic Escherichia coli and detection of traT gene in isolated from patients referred to Abadan hospitals during 2017-2018
Objective: Escherichia coli is one of the most important causes agents of urinary tract infection in human. Thus, identification of Escherichia coli resistance patterns seems to be necessary. traT gene has been reported variable in Escherichia coli strains isolated from urinary tract infection. Therefore, the aim of this study was to investigate the prevalence of uropathogenic Escherichia coli ...
متن کاملSensitivity of Escherichia coli to viral nucleic acid, X. Ba2+-induced competence for transfecting DNA.
Effect of alkaline earth metal ions on induction of the competence for DNA transfection was investigated. Unlike spheroplasts, the bulk of the bacteria treated with these ions retains colony-forming ability. The order of effectiveness for transfection of phiA replicative-form DNA has been found to be Ba2+ greater than Ca2+ greater than Sr2+ greater than Mg2+. The competence of Ba2+-treated cell...
